The present invention relates to a pharmaceutical composition for preventing or treating micro-RNA-34a (miRNA-34a) related diseases comprising an expression promoter or an activator of TIS21 as active ingredients; and a therapeutic screening method thereof. More specifically, when a pharmaceutical composition according to the present invention is administered in a p53 mutant cell in which p53 expression of miRNA-34a whose expression is regulated by p53, is inhibited, it is confirmed that expression of TIS21 is induced and expression induced TIS21 induces expression of miRNA-34a in a p53 mutant cell. Thus, a pharmaceutical composition comprising an expression promoter or an activator of TIS21 as active ingredients can be used as a pharmaceutical composition for preventing or treating miRNA-34a related diseases. In addition, in a tissue of a patient having miRNA-34a related diseases in which expression of miRNA-34a is reduced, by using a screening method of an expression promoter or an activator of TIS21, drugs for promoting or activating TIS21 expression can be specifically selected.
