The present invention relates to a manufacturing method of bio-oil which has a high concentration of DHA through culturing Thraustochytrid-based microalgae using saccharification liquid of a fiber-based palm-oil industry byproduct. More particularly, the manufacturing method of the present invention produces bio-oil by culturing Thraustochytrid-based microalgae in a culture medium with saccharification liquid of a fiber-based palm-oil industry byproduct, and recovers the generated bio-oil. The manufacturing method of bio-oil according to the present invention can produce bio-oil from a non-edible fiber-based biomass rich in nutrients through culturing Thraustochytrid-based microalgae, and thus can surpass limitations in developing bio-fuel, such as unstable supply and demand of food resources, increasing price of raw materials, etc., while securing commercial competitiveness of microorganism fermented oil.;COPYRIGHT KIPO 2016
