Provided are a collagen sponge containing fracture healing promotion drug and preparation method thereof. The preparation method comprises the following steps: 1) adding, to 1 kg of an enzymatic hydrolysate of collagen, a bisphosphonate containing a phosphonate equivalent to a 70-140 mg dose of phosphonate and/or 100-800 mg of a strontium salt, wherein the ratio of collagen hydrolysates in the enzymatic hydrolysate of collagen to water is 1:9-1:8; 2) adding a cross-linking agent to the enzymatic hydrolysate of collagen containing the bisphosphonates and/or the strontium salt at a proportion of 40-60 U of the cross-linking agent per gram of the enzymatic hydrolysate of collagen, and performing cross-linking for 2-5 hours, wherein the cross-linking agent is a transglutaminase; 3) evenly stirring a mixed liquid acquired in step 2), placing the mixed liquid in a stainless steel plate, placing the stainless steel plate in a vacuum freeze dryer, and freeze-drying the mixed liquid for 10-16 hours to transform the same into a spongy state; and 4) cutting, packing, and sealing the collagen sponge containing the bisphosphonates or the strontium salt and acquired in step 3), and exposing the same to Cobalt 60 radiation for sterilization.
