The present invention relates to a method for simultaneously detecting multiple diseases based on a high-sensitivity 3D protein nanoparticle probe, and to a kit for simultaneously detecting multiple diseases by using a high-sensitivity 3D protein nanoparticle probe. Specifically, the present invention relates to a lateral flow assay kit comprising: a sample pad; a conjugation pad in which a signal molecule is planted; a membrane which includes a protein nanoparticle probe in which a disease marker-specific epitope is fused and expressed on the surface of a self-assembly protein; and an absorption pad, and to a method for simultaneously detecting multiple diseases by using the kit. The protein nanoparticle probe according to the present invention has disease marker-specific epitopes expressed with correct orientation and high density on its surface, and by adjusting and varying their mixing ratio, can detect various disease marker-specific epitopes. Further, a disease marker detection method using such probes shows superior sensitivity and accuracy owing to having various disease marker-specific epitopes expressed on the surface of the protein nanoparticle.
