The present invention relates to the stratification of epithelial-mesenchymal transition gradients in three-dimensional mesenchymal cell spheres. In the present invention, it was found that the changes in the nitric activity of mesenchymal stem cells are closely related to the epithelial-mesenchymal transition. Specifically, when the niacin activity was enhanced by 3D-cell interactions or related physiological stimulation, the gradient of epithelial-mesenchymal transition was further changed to mesenchymal state, and the opposite tendency was observed in the degenerative state caused by leukemia . Thus, the present invention is expected to improve the efficacy of mesenchymal stem cell-based cell therapy by modulating or introducing epithelial-mesenchymal transition-related factors.
