The present invention relates to hierarchy of epithelial to mesenchymal transition gradient in three-dimensional spheroid mesenchymal stromal cells. It was discovered in the present invention that variations in the niche activity of mesenchymal stem cells are closely correlated with epithelial to mesenchymal transition. In detail, an improvement in niche activity by interaction between 3D cells or by related physiological stimuli further shifts an epithelial to mesenchymal transition gradient toward a mesenchymal state while reverse tendency is observed in a leukemia-caused degeneration state. Therefore, the present invention is expected to enhance the efficiency of mesenchymal stem cell-based cell therapy by controlling or introducing factors responsible for epithelial to mesenchymal transition.
