The present invention provides an aqueous tumor-directed liposome nanoparticle composition comprising an aqueous dispersion of liposome nanoparticles. the nanoparticles preferably encapsulate an anti-tumor chemotherapeutic agent which may be added to a preformed liposome composition or may be incorporated into the liposomes during liposome formation. liposome nanoparticles comprise a legumain-directed lipid in admixture with one or more other micelles or lipid vesicle-forming materials in the form of liposome nanoparticles dispersed in an aqueous vehicle. A preferred tumor-directed liposome nanoparticle composition comprises (a) a legumain-directed lipid component, (b) a zwitterionic lipid component, (c) an amino-substituted lipid component, (d) a neutral lipid component, and (e) polyethylene glycol conjugate lipid component. the legumain-directed lipid component comprising a hydrophobic lipid moiety covalently linked to a legumain binding moiety.
