The present invention relates to a method of manufacturing a shiitake beef jerky using jujube, and more specifically to easily provide access to a mixture of shiitake mushroom and beef jerky in everyday life by: removing the skin of shiitake mushrooms and cutting the shiitake mushrooms with a knife at a predetermined interval, and shredding the cut pieces of the mushroom as well as beef meat for jerky to form pulverized beef; mixing the pulverized shiitake mushroom and beef, inserting and stirring honey and pine needle powder with the same; naturally drying the mixed body to cover the exterior of jujube after seeds have been removed by preparing a plate-like shiitake mushroom jerky to cover the exterior or jujube; rolling up the cover of the jujube after the seeds have been removed with the shiitake jerky; and preparing the jujube without seeds by naturally drying and maturing the shiitake and beef jerky. According to the same, vitamin D, high content of protein, and cholesterol reduction, which are the effects of shiitake mushrooms against adult diseases, and the taste of shiitake mushroom which increases the flavor, as well as the tranquilizing effect and restoration of stamina, which are the effects of jujube can be easily experienced by eating the shiitake beef jerky.
