The present invention relates to a fatty acid quantifying method, and more specifically, to an improved method for simultaneously quantifying fatty acids in a driven blood spot (DBS) with LC/ESI-MS/MS by picolylamine derivatization. An analysis method according to the present invention is advantageous in that storage and sample stability are superior than the conventional blood specimen by using the DBS as a specimen to be analyzed. Also, the analysis method of the present invention is advantageous as compared with the conventional analysis method using blood in that using the DBS as a specimen causes less stress than blood collection and enables easy sample collection by an ordinary person, thereby being used as a point-of-care test kit.
