The present application relates to 3,5-Diarylidenyl-N-substituted-piperid-4-one analogs, and pharmaceutically acceptable derivatives thereof, which are useful in the treatment or prevention of disorders comprising cancer, autoimmune disorders, inflammatory disorders, and fibrotic disorders; to pharmaceutical compositions comprising the compounds; to methods of treating disorders, such as cancer, associated with aberrant Stat3 pathway activity by using the compounds, and compositions further comprising another therapeutic agent, such as an anticancer drug; to methods of using such compounds or compositions thereof to treat resistant, and/or metastatic cancers; to methods of inhibiting Stat3 pathway activity in a cell; to methods for making the pharmaceutical compounds; to synthetic methods for making the compounds; and to compounds and compositions useful as a fluorescent probe.
