Disclosed is a quantitative detection method of a glucose level based on an RF patch biosensor combined with volume-fixed structures. The method proposed a characterization concept of volume-fixed structures and an RF patch biosensor in order to monitor individual′s state of glucose levels of a diabetic patient. The patch biosensors of two types in which a rear slot (0.53 μL) and a front tank (0.70 μL) are individually integrated have been developed in order to accurately and effectively detect a glucose test sample except interference of liquid states, shapes and thicknesses. Glucose which is a test material with various concentrations of 25 mg/dL to 1,000 mg/dL is dropped onto volume-fixed structures in various concentrations, and effectively interacts with an electromagnetic field of a wireless frequency patch to sensitively change resonant frequencies and sizes. Measurement of reflection coefficient was performed in a probe station to prove that two types of patch biosensors can detect the glucose level, and the two biosensors show high sensitivity of 1.13 MHz to 1.97 MHz per 1 mg/dL, and show a short response time of less than one second and a low detection limit of 26.54 mg/dL and 15.22 mg/dL.
