The present invention relates to a nucleic acid molecule capable of expressing a gene of interest using an expression control sequence derived from a virus or eukaryotic cell having an internal ribosomal binding site (IRES), an expression vector into which the nucleic acid molecule is inserted, and an application thereof. The nucleic acid molecules of the invention can be used to efficiently express a gene of interest without the complex capping process associated with expression of the gene of interest. According to the present invention, a nucleic acid molecule having a plurality of adenosine or thymine linked to an expression control sequence can be used to efficiently express a gene of interest. By applying an expression system having a nucleic acid molecule according to the present invention, it is expected to be used as a gene vaccine or as a gene therapy agent for expressing a protein or peptide related to disease treatment.
