A method and kit for rapid nucleic acid extraction from nucleic acid containing materials in bone, dental or semen samples are disclosed.For the bones and teeth, the method comprises providing a nucleic acid containing material in the form suitable for nucleic acid extraction, adding a dissolution buffer to the nucleic acid containing material to obtain a mixture, mixing the mixture by about 30 seconds or more by an equivalent method, and then separating the mixture by centrifugation to obtain a liquid supernatant.Liquid supernatant contains extracted nucleic acids that can be used for analysis involving STR profiling by conventional or rapid DNA analysis.For the semen, the method and kit include applying an appropriate amount of sperm destructant.
