一种纤维二糖差向异构酶突变体及其在制备乳果糖中的应用

摘要

本发明公开了一种纤维二糖差向异构酶突变体及其在催化法制备乳果糖中的应用。所属突变体是对氨基酸序列如SEQ ID NO:1所示的纤维二糖差向异构酶CE进行单点突变或者组合突变,得到了催化活性提高的纤维二糖差向异构酶突变体,可用于以乳糖为底物异构化生产乳果糖。与野生型纤维二糖差向异构酶相比,其单点突变体和组合突变体活性更高,尤其是组合突变体,表现出单点突变体活性的叠加效果,突变体R56K/L187I/L254I粗酶液酶活高达30U/ml。在80℃、pH7.0、底物浓度700g/L的反应条件下,其催化乳糖生成乳果糖的产量为526g/L,转化率达75%,大大降低了生产成本。

技术领域

本发明属于生物技术领域,涉及一种纤维二糖差向异构酶突变体的应用,具体涉及一种纤维二糖差向异构酶突变体在以乳糖为底物异构化反应制备乳果糖中的应用。

背景技术

乳果糖化学名4-O-β-D-吡喃半乳糖基-D-果糖,与乳糖是同分异构体,甜度相当于乳糖小于蔗糖,带有清凉醇和的感觉,粘度低,热值低,安全性高,稳定性好,不发生美拉德反应。乳果糖具有调节肠道菌群、提高免疫力、改善新陈代谢等独特生理功能,还可以降低血氨、导泻,用于各种肝性疾病的防治,在食品方面可以做为低热值甜味剂和功能性食品添加剂,是一种药食两用的功能性低聚糖。

目前商业化生产乳果糖主要是乳糖在碱性溶液中通过化学异构化获得,需要大量的化学催化剂而且反应条件一般比较激烈,得到的产品色泽深,副产物多,且催化剂分离困难,制约了化学异构法生产乳果糖的进一步发展。因此,酶法合成乳果糖是一种经济有效的替代方式,乳糖经半乳糖酰化或异构化可有效地生产乳果糖。其中,半乳糖酰化通常是通过β-葡萄糖苷酶和β-半乳糖苷酶(EC 3.2.1.23)催化乳糖水解产生半乳糖和葡萄糖,半乳糖部分转化为果糖,然后果糖与半乳糖在转糖苷酶的作用下形成乳果糖;纤维二糖-差向异构酶则能催化乳糖异构化生产乳果糖。

β-葡萄糖苷酶:Fischer等在1996年发现产自Pyrococcus furiosus的CelB酶具有100%的β-葡萄糖苷酶和60%的β-半乳糖苷酶活性。2004,Mayer 等[1]研究发现,在果糖作为半乳糖基受体的条件下,重组CelB催化合成乳果糖的最高产量为15.0g/L;2010年,该研究小组开发了一种在酶膜反应器或填充床上进行连续化生成乳果糖的模型,由固定在阴离子交换树脂Amberlite IRA-93 或Eupergit® C上的CelB作为催化剂合成的乳果糖产量均为14.6g/l,而由游离型CelB转化得到的乳果糖产量则为13.9g/l[2]。

游离β-半乳糖苷酶:2006,Doek等[3],采用基因修改耐热型乳糖酶,以乳糖(400g/L)和果糖(200g/L)为底物,在80℃反应6h产生了50g/l 的乳果糖;2008,张萃荟等[4],采用乳酸克鲁维酵母,经过诱变后筛选出的菌株,β-半乳糖苷酶酶活约为33.47U/mL,乳糖和果糖的最佳浓度分别为40%(w/v)和20%(w/v),在最佳反应条件下乳果糖的产量为57.83g/;2009,Adamczak[5]发现米曲霉β-半乳糖苷酶催化乳果糖合成的能力最强,产量最高达65g/L;2011,Tan等[6]发现低温节杆菌(Arthrobacter sp.)β-半乳糖苷酶在温和的反应条件(20℃,p H 6.0)下也可以催化生成一定的乳果糖。

固定化β-半乳糖苷酶:2010,Hua X等[7],利用固定化的β-半乳糖苷酶和葡萄糖异构酶,在环己烷-缓冲液双相体系中,乳果糖的最高产量可达151 g/L,得率达到17%;2015,沈秋云等[8],将包埋于海藻酸盐-明胶-磷酸钙(AGCaP)胶囊的β-半乳糖苷酶连续使用20次以后,酶活仍保持60%以上,存放30天以后,酶活仍然保持在90%左右。同时,与游离的及包埋于海藻酸钙胶囊的β-半乳糖苷酶相比,固定于AGCaP的β-半乳糖苷酶具有最广的温度和pH适用范围。38℃,pH 6.8,乳糖和果糖浓度分别为250 mg/mL条件下,反应3 h后,包埋于AGCaP胶囊的β-半乳糖苷酶催化转糖苷反应分别生成26.1 mg/mL乳果糖和74.2 mg/m L乳果糖。2018,Guerrero C 等[9]采用固定化米曲霉来源的β-半乳糖苷酶以果糖和乳糖为原料连续生产乳果糖,乳果糖的最高等率达到0.6 g/g。2020,Maria de Fátima Matos deFreitas1等[10],β-半乳糖苷酶是由乳酸克鲁维酵母 NRRL Y1564在最适温度30℃的干酪乳清中添加酵母提取物产生的,其酶促活性为4418.37 U / g。通过可溶性和固定化酶两者水解乳糖,获得的转化率分别为38.0%和42.8%,使用从固定化酶到壳聚糖载体的生物催化剂,乳果糖的产量也可以达到17.32 g / L。

以上介绍了传统酶法制备乳果糖主要的3种形式,虽然众多研究学者对传统酶法制备乳果糖进行了各种改进,包括有机双水相体系降低水分活度,从而提高β-半乳糖苷酶的转糖苷活力、采用双酶共固定化体系催化单一底物乳糖反应以及透性化细胞的使用等,但是传统酶法制备乳果糖仍存在转化率低(10%~43%)、共同作用底物果糖浓度高(100g/L~300g/L)、反应时间长、生产效率低(≤151 g/L)等缺陷,限制了传统酶法在乳果糖制备中的大规模应用。

纤维二糖差向异构酶是一种嗜热酶,在高温下底物溶解度、反应速率都会提高,应用纤维二糖差向异构酶的好处在于它不需要额外添加底物果糖,成本低廉,所以未来利用它进行商业化生产乳果糖的前景十分广阔。2012,Yeong-Su等[11]尝试用来源于Caldicellulosiruptor saccharolyticusDSM 890的能将醛糖异构化为酮糖的纤维二糖差向异构酶(CsCE)催化乳糖反应,在最适条件(pH 7.5,80℃,700g/l乳糖)下,乳果糖产量可达408g/l,乳果糖的转化率为58%,同时有约15%的底物乳糖转化为依匹乳糖,乳果糖的浓度和转化率都是酶法制备乳果糖中最高的报道;2013,该Yeong-Su研究组[12]在CsCE催化乳果糖合成的反应体系中加入硼酸盐(摩尔比,硼酸盐:乳糖=1:1),乳果糖的产量提高到616g/l,转化率提高到88%,虽然这种方法可以提高乳果糖转化率,但是大量硼酸盐的加入,提高了分离纯化的难度,增加了乳果糖合成的成本。专利CN201910023301.9公开了一种包含纤维二糖差向异构酶突变体的重组枯草芽抱杆菌,重组差向异构酶酶活达8U/mL,在80℃、pH 7.5 、加酶量20U/mL 、底物浓度400g/L的反应条件生成乳果糖的产量为200g/L,转化率可达51%。

综上所述,现有传统酶法及纤维二糖差向异构酶法制备乳果糖的技术任然存才底物得率低、成本高等问题,因此,需要寻找新的活性更高、稳定性更好的维二糖差向异构酶。

发明内容

本发明所要解决的技术问题是提供一种纤维二糖差向异构酶突变体,及其在催化乳糖异构化制备乳果糖中的应用。

为解决上述技术问题,本发明所采用的技术书方案为:

本发明提供一种纤维二糖差向异构酶突变体,所述纤维二糖差向异构酶突变体是在SEQ IDNO :1所示的氨基酸序列上进行突变,所述突变位点选自R56K、L187I、L254I、L254M、W372Y、C374T中一种或一种以上组合。

优选地,该纤维二糖差向异构酶突变体,所述突变位点为R56K、L254I、C374T、R56K/ W372Y、R56K/ L254M、L254I/C374T、R56K/L187I/L254I、R56K/L254I/C374T或R56K/L254I/W372Y。

进一步优选的,所述突变位点为R56K/L187I/L254I或者R56K/L254I/W372Y。

本发明提供的纤维二糖差向异构酶突变体,R56K对应的单点突变体的氨基酸序列为SEQ ID NO :3;L254I对应的单点突变体的氨基酸序列为SEQ ID NO:5;C374T对应的单点突变体的氨基酸序列为SEQ ID NO :7;R56K/ W372Y对应的组合突变体的氨基酸序列为SEQID NO :9;R56K/ L254M对应的组合突变体的氨基酸序列为SEQ ID NO :11;L254I/ C374T对应的组合突变体的氨基酸序列为SEQ ID NO :13;R56K/L187I/L254I对应的组合突变体的氨基酸序列为SEQ ID NO :15;R56K/L254I/C374T对应的组合突变体的氨基酸序列为SEQID NO :17;R56K/L254I/W372Y对应的组合突变体的氨基酸序列为SEQ ID NO :19。

本发明还提供一种编码如上所述的纤维二糖差向异构酶突变体的基因,

本发明还提供一种包含如上所述的基因的重组表达载体。本发明还提供一种包含如上所述的纤维二糖差向异构酶突变体的可溶性蛋白、固定化酶或工程菌。

所述纤维二糖差向异构酶突变体在催化乳糖异构化制备乳果糖中的应用。

本发明还提供一种生产乳果糖的方法,以乳糖为底物,以如上所述纤维二糖差向异构酶突变体为催化剂。

优选地,如上所述生产乳果糖的方法为,乳糖浓度500-700g/L,加酶量20-30U/mL,反应温度75-80℃,pH为7 .0-7 .5,反应时间为1-4h。

本发明还提供如上所述纤维二糖差向异构酶突变体在食品、饲料或医药领域中的应用。

在食品领域,乳果糖可以刺激成年人及未成年人肠道中益生菌的生长,基于这种有助健康的特性,可作为功能食品中的理想营养添加源;一些为特定疾病(如糖尿病)的病人准备的特殊功能性食品中也可以添加乳果糖;乳果糖作为乳制品的添加剂,例如乳果糖酸奶可用于治疗儿童便秘,相比于含有其他膳食纤维(如低聚半乳糖、抗性淀粉等)的酸奶,会有更好的治疗效果。在饲料领域,乳果糖可作为动物饲料增加家畜的机体免疫力。在医药领域,乳果糖具有抑制沙门氏菌,降低血氨值,增强矿物质吸收,阻止胆汁酸形等作用,可以用于治疗便秘,肝性脑病,肠炎疾病,还用于渗透压调节类泻药。

有益效果:本发明首次对氨基酸序列如SEQ ID NO :1 所示的纤维二糖差向异构酶进行单点突变或组合突变并应用于催化制备乳果糖,催化活性显著提高,其中组合变体R56K/L187I/L254I粗酶液酶活高达30U/ml,在80℃、pH7.0、底物浓度700g/L的反应条件下,催化乳糖生成乳果糖的产量为526g/L,转化率达75%,大大降低了生产成本。

具体实施方式

HPLC法测定乳果糖:戴安U3000,BP-100钙离子碳氢化合物柱和RI检测器,柱温80℃,流动相为超纯水,流速0.5 mL/min。采用外标法,根据峰的保留时间和峰面积来确定乳果糖的产量。

纤维二糖差向异构酶酶活的定义:在80℃,pH7.0,50mM缓冲液中,每分钟催化乳糖生成1μmol 乳果糖所需的酶量定义为一个U,反应底物乳糖浓度为150g/L,反应时间20min。实施例1:重组质粒pET-28a-CE的转化

由通用生物系统(安徽)有限公司合成得到包含氨基酸序列如SEQ ID NO :1 所示的纤维二糖差向异构酶CE(GenBank: HGB31345.1)基因的重组质粒pET-28a-CE,将该重组质粒转化至购买于诺唯赞的大肠杆菌E.coli BL21感受态中,得到重组大肠杆菌E.coliBL21 (DE3)/pET-28a-CE,涂布含卡那霉素(50μg/mL)抗性的LB平板上,37℃过夜培养。挑选单菌落至含卡那霉素(50μg/mL)抗性的5mL液体LB培养基中,37℃培养8h后,菌液保存甘油管,贮存于-20℃冰箱,经通用生物测序验证正确后进行摇瓶发酵产酶。

实施例2:突变体的获得

采用定点突变技术获得单点突变或组合突变体,定点突变的引物采用Primerpremier 5.0 设计,引物设计的原则为:正反向扩增引物5’端包含15-21bp反向互补区域,各引物非互补区域长度至少为15 bp,所需引入的突变包含在互补区域内。突变引物见表1。

定点突变以pET-28a-CE重组质粒为模板,利用PrimerStar Mix进行全质粒扩增,按照表2设置反应体系,扩增产物经过Dpn I酶消化去除PCR反应体系中的模板,然后在重组酶催化下将5’端和3’端进行同源重组,完成质粒的环化。最后将环化的扩增产物转入大肠杆菌BL21(DE3)宿主菌,并涂布在含卡那霉素的平板上,置于37°C培养箱中过夜培养。

PCR程序为:95°C预变性300s,98°C变性10s,65°C退火15s,72°C延伸300s,反应30个循环后,再72°C延伸5min,最后4°C保温。PCR反应结束后,用0.8%的琼脂糖凝胶电泳检测PCR产物。然向每个PCR管中加入1µl Dpn I 轻轻混匀后置37°C金属浴2 h,之后将消化好的扩增产物按照表3中的配方进行重组反应。

次日从平板中挑选含有突变质粒的重组大肠杆菌 BL21三株,将上述重组菌从平板分别接种到装有5 mL含有相应抗性的液体LB培养基(LB (g/L):蛋白胨 10,氯化钠10,酵母提取物5)的50mL的摇管中,加入相应抗性,在摇床上37°C恒温培养12 h,转速200 rpm。培养结束后抽提质粒,送金斯瑞公司进行测序。最后将测序结果与野生型酶蛋白核酸序列进行比对,确定突变是否成功。

按照上述方法先进行单个位点突变依次得到重组表达菌E.coli BL21 (DE3)/pET-28a-R56K、E.coli BL21 (DE3)/pET-28aL187I、E.coli BL21 (DE3)/pET-28a-L254I、E.coli BL21 (DE3)/pET-28a-L254M,E.coli BL21 (DE3)/pET-28a-W372Y、E.coli BL21(DE3)/pET-28a-C374T,再以进行过一次突变后的重组质粒为模板进行二次或三次突变,最后得到了测序正确的包含CE突变体的双位点以及三位点突变重组表达菌E.coli BL21(DE3)/pET-28a-R56K/W372Y、E.coli BL21 (DE3)/pET-28a-R56K/L254M、E.coli BL21(DE3)/pET-28a-L254I/C374T、E.coli BL21 (DE3)/pET-28a-R56K/L187I/L254I、E.coliBL21 (DE3)/pET-28a-R56K/L254I/C374T、E.coli BL21 (DE3)/pET-28a-R56K/L254I/W372Y。

实施例3:重组菌大肠杆菌发酵培养

将实施例1、2中得到的10株(不含E.coli BL21 (DE3)/pET-28a-L187I,E.coliBL21 (DE3)/pET-28a-L254M,E.coli BL21 (DE3)/pET-28a-W372Y)重组菌大肠杆菌菌株分别接种于5ml含卡那霉素(50μg/mL)抗性的LB培养基中,37℃下培养8h后以2%接种量转接至50mL TB发酵培养基中,先于37℃、200rpm恒温培养至菌体OD

实施例4:重组酶CE及突变体酶催化

反应体系为:乳糖浓度500g/L,加酶量20U/mL(重组酶CE或突变体酶),使用pH7.0的磷酸钠缓冲,在75℃、150r/min水浴摇床中反应,每隔1h取样煮沸终止反应,用HPLC检测产物的含量,4h后终止整个反应。

酶催化结果如表5所示,经检测当反应进行到3h,重组酶CE达到平衡转化率56%,而突变体酶R56K/L187I/L254I转化率高达75%。

实施例5:一种生产乳果糖的方法

以乳糖为底物,以纤维二糖差向异构酶突变体R56K/W372Y,R56K/L187I/L254I或R56K/L254I/W372Y为催化剂,反应条件:乳糖浓度700g/L,加酶量30U/mL,反应温度80℃,pH为7.5,150r/min水浴摇床中反应,每隔1h取样煮沸终止反应,用HPLC检测产物的含量,直至反应达到平衡。

酶转化反应过程如图2所示,当反应进行到3h,突变体酶反应均达到平衡,且R56K/L187I/L254I转化率最高达75%,产量为526g/L,其液相色谱峰图如图3所示。
