8-Chloroadenosine-3',5'-Cyclic Monophosphate (NSC-614491) Pellet Implantation Study in Female Nude Mice.

NCr-nu mice were subcutaneously implanted with 8-chloro-cyclic adenosine monophosphate/cholesterol pellets on study days 1, 8, 15, 22, 29, 36, 43, and 50 at doses of 0.0, 0.1, 1.00, and 3.00 mg/pellet/animal revealed no alterations in serum blood urea nitrogen and creatinine values. No drug-related gross lesions were observed from animals necropsied on study days 16, 33, 47, or 54. In addition, histopathologic examination of the kidneys or tissues/organs associated with the gastrointestional tract detected no drug-related lesions or birefringent material in unstained frozen kidney sections. Little of the drug was absorbed from the pellets into the plasma and an endogenous peak chromatographing at the same retention time as the drug made quantitation uncertain.
