Sulphur Mustard Inhibits Heat Shock Induction of HSP70 in Human Keratinocytes

The effect of sulfur mustard (SM) on induction of the 70kDa heat shock protein(HSP70) in a human keratinocyte cell line has been investigated. Westernblots stained with a monoclonal anti-HSP70 antibody, or incorporation of 35S-methionine into the HSp70 band in SDS-PAGE gels, showed that exposure of cells to SM inhibited the heat shock induction of HSP70. A monofunctional analogue of SM, 2-chloroethyl ethylsulphide (CEES) inhibited HSP70 synthesis in the same manner, showing that the crossbuilding activity of SM is not needed for inhibition. Sodiumsalycylate, which induces binding of heat shock factor to heat shock element, did not protect against the inhibition. Since skin is one of the major targets for damage upon exposure to SM, the inhibition of HSP70 synthesis might have influence on the severity of damage.
