Toxic Effects of the Daily 1.M Injection of Pregn-4-Ene-3, 11, 20-Trione 6 Alpha-Methyl-(Nsc-17 256E) IN Dogs

NSC-17 256E, Ben Venue Labs. Lot 68-218, dissolved (30 mg/ml) in cottonseed oil with 5% Benzyl alcohol and 15% Benzyl benzoate was injected 1.M. in a total of 8 dogs. Two other dogs were treated with the vehicle alone. All dogs survived. The ones treated with large doses of NSC-17 256E reacted with reversible abdominal tenderness, occasional diarrhea, and sometimes painful swelling at the injection sites, but were otherwise in good general condition. Treatment with 5 consecutive daily doses of 30 mg/kg, with 15 consecutive daily doses of 15 or 5 mg/kg elicited dose-related moderate loss of weight, marked to moderate neutrophilia, moderate lymphopenia and often complete absence of the eosinophils in the peripheral blood. There also occured marked but reversible derangement of the hepatic tests, and a decrease in serum levels of K and Cl. The autopsy at the end of the treatment period revealed increased size of the liver, severe fat and glycogen deposits in the hepatocytes, hypocellularity of the lymphoid tissues, toxic changes of the adrenal cortex, stimulation of the female reproductive tract, degenerative changes of the prostate gland and inflammation at the injection sites. These changes indicate glucocorticoid, thymolytic anti-ACTH and slight progestational activity. (Author)
