A method has been developed for the analysis of Dapsone (DDS) and 4'-((4-aminophenyl)sulfonyl)acetanilide) (MADDS) which is based on converting them to 4-iodophenyl sulfone and 4-amino-4'-iodophenyl sulfone, respectively. These compounds are then separated by gas chromatography and measured by means of an electron capture detector. Sensitivity is at the 1 - 10 ng/ml level and no interferences have been found thus far in human plasma. A method was investigated for the measurement of DDS, MADDS, and 4',4''-sulfonyldiacetanilide (DADDS) at the 100 nanogram level by reducing them with lithium aluminum hydride and measuring the reduction products by gas chromatography. The fluorescence characteristics of the compounds obtained on reducing MADDS and DADDS were investigated. It was found that monethyl-DDS (from MADDS) and diethyl-DDS (from DADDS) are more highly fluorescent than their precursors. Preliminary investigations were undertaken to assess the value of various methods for the analysis of B663. This compound does not fluoresce but the electron capture detector is highly sensitive to it. (Author)
