Toxicity Studies on Compound Nsc 102627 Following Intravenous Administration to Mice and Beagle Dogs

The toxicity of compound NSC 102627 formulated in physiological saline was studied following single and five day repeated I.V. dosages to make and female mice and following I.V. administration to beagle dogs. The drug was more toxic to mice when given in multiple doses than following single administration, and when given in a single dose it was more toxic to the male mice than the females. Toxic manifestations included weight loss, diarrhea, 'seedy appearance' and in a few of the female animals, tremors. The majority of deaths noted on both dosage schedules occurred during the first seven days with occasional delayed deaths. In the beagle dogs single I.V. doses of 40 and 15 mg/kg were lethal to the animals and the dose of 7.5 mg/kg was toxic but tolerated. Toxic signs noted consisted of weight loss, anorexia, hyperthermia, leucopenia with a decrease in granulocytes, a slight anemia, BSP retention on the higher dosed animals and an increase in alkaline phosphatase in the animal given 15 mg/kg.
