Toxicity of A and UI Formulations of Ellipticine Following a Single Oral Dose to Beagles

The toxicity of single oral doses of Ellipticine A and UI formulations were evaluated in beagle dogs. The objective of this study was to determine oral dose levels that were non-emetic to beagles. Criteria evaluated for toxic effects included clinical observations, clinical laboratory studies, necropsy findings and microscopic pathology. Evidence of drug-induced toxicity was limited to the appearance of emesis in the pairs of animals receiving 200 mg/cu m of Ellipticine of either the A or UI formulations. Emesis was observed at 3 hours posttreatment with the Ellipticine A and at 2 and 3 hours posttreatment with the Ellipticine UI formulations. No drug-related toxic effects were observed in the pairs of animals receiving either formulation at the 100 mg/cu m (5 mg/kg bw) level.
