2,2-Bis(Bromomethyl)-1,3-Propanediol: Reproduction and Fertility Assessment in CD-1 Mice When Administered in Feed (Revised January 1986)

2,2-Bis(bromomethyl)-1,3-propanediol (BBMP) is used as a flame retardant for epoxy, polyester, and urethane foams. The route of administration was feed. Rats ingesting 5 mg/kg bw of BBMP showed no symptoms of toxicity. Treatment at 100 mg/kg resulted in significant toxicity, including degenerative changes in the liver, eyes, and possibly the thyroid gland. BBMP administered in feed at the 0.4% dose level adversely affected reproduction in CD-1 mice. The continued BBMP treatment at this dose level also resulted in a significant drop in body weight. When female mice exposed to 0.4% BBMP were mated with control male mice (Task 3), the fertility index was reduced by almost 60%. The number of live pups per litter delivered by these breeding pairs was also significantly lower than the 0.4% male X control female pairs.
