Since Sanger sequencing was first introduced as a rapid method of DNA sequencing in the early 1970s, many innovative technologies have been developed to provide ever-higher throughput. DNA microarrays have become the most widely used systems for interrogation of large segments of DNA. This technique involves fragmenting and labeling the DNA sequence of interest, followed by hybridization to an array of oligonucleotide probes of known sequences, and subsequent detection and analysis. However, this system has a limited dynamic range, and can suffer from sensitivity, reproducibility and specificity issues.
