Polymerase chain reaction (PCR) and restriction fragment length polymorphism (RFLP) analysis of toll-like receptor 4 (TLR-4) gene was performed on blood samples from 102 Murrah Buffaloes. Amplified PCR fragment of TLR-4 exon 1, exon 2, exon 3.1 and exon 3.2 RFLP produced fragments of 165, 300, 478 and 409 bp, respectively. The PCR-RFLP analysis using Dra I, Hae III and Hinf I restriction enzymes revealed polymorphisms in exon 1, exon 3.1 and exon 3.2 of the TLR 4 gene. Hind III did not cut any sequence. Genotypes frequencies were 0.088-0.676 for AA, 0.078-0.746 for AB and 0.049-1.000 for BB genotypes. The B allelic frequency (0.186-1.000) was more prevalent than allele A (0.814-0.206). Results suggest that polymorphisms in TLR-4 gene could be used as a possible marker for identifying mastitis resistance in Murrah buffalo.
