Progesterone (P) regulates female reproduction via two nuclear receptors, PR-A and PR-B. Although both receptors display overlapping and distinct transcription regulatory properties; PRs function as transcriptional mediators to regulate target gene transcription and affect the neural networks involved in the control of female reproductive behaviour. Single nucleotide polymorphisms (SNPs) represent a potential resource for analysing DNA sequence variation in many animal species. SNPs have become the markers of choice in genetic studies because of their high frequency, low mutation rates and amenability to automated analysis. Genomic DNA extracted from10 healthy multiparous female buffaloes was amplified using primers that were designed from the cattle PR-A gene sequence. The amplified fragments of PR-A (560-bp) were digested with Dra restriction enzyme. Sequence alignment was achieved by Codon Code Aligner 5.0.1 and analysis was carried out using NCBI/BLAST/blastn suite. The results showed that alltested buffaloes showed the one kind of digestion pattern, a finding characterizing the region as monomorphic. The study of other regions of PR-A gene might provide molecular data that can be used in the future for the selection of reproductive animals.Genetic improvement of buffalo productivity has been dependent on concepts of quantitative genetics of various productivity traits of economic importance to promote more efficient and relatively easy selection of Indian buffaloes.
