Chronic hypoxia (24 hours, 1% O_2) was shown to decrease mRNA expression of neprilysin (NEP) and insulin-degrading enzyme in human neuroblastoma NB7 cells by 25% and 33%, respectively. Addition of vilon (50 nM) to the culture medium did not significantly change mRNA expression of NEP after hypoxia, while epithalon in the same concentration induced an increase in the expression by 28% in comparison with the control level. Epithalon was also able to completely prevent downregulation of the expression of the insulin-degrading enzyme during hypoxia. The observed effects of epithalon on neprilysin were comparable to those of the antioxidant epigallocathechin-3-gallate.
