Re: Combined Androgen Deprivation Therapy and Radiation Therapy for Locally Advanced Prostate Cancer: A Randomised, Phase 3 Trial

The study by Warde et al. had the objective of determining the importance of local control in locally advanced prostate cancer (PCa) treated with androgen deprivation therapy (ADT). Between 1995 and 2005, >1200 patients with locally advanced/ high-risk PCa were included in the study. Patients had T3/T4 cancers or T2 cancers with high-risk features such as very high prostate-specific antigen levels and/or Gleason scores of 8-10. All patients received lifelong ADT and were randomized to receive either radiation therapy (RT) or no additional therapy. The results confirmed that the combination of ADT plus RT was superior to ADT alone in terms of progression-free survival [PFS], cancer-specific survival, and overall survival. These differences were all statistically significantly. The added grade 3 toxicity of RT was low, and the combination was generally well tolerated.
