Production of beta-galactosidase using novel yeast isolate from whey.

The hydrolysis of lactose by the enzyme beta-galactosidase into glucose and galactose plays an important role in the biotechnology, pharmaceutical and food processing industries. This study focused on the isolation of novel yeast strains and the formulation of a low-cost culture medium based on whey for optimal beta-galactosidase activity by the isolated yeast strains. The yeast isolate identified as Kluyveromyces marxianus showed maximum enzyme activity (1710 IU/g DW) when grown on whey permeate at pH 5.5, 30 degrees C, after an incubation period of 28 h. Utilization of the dairy industry waste whey was found to be economical for beta-galactosidase production by the isolated yeast cells. This enzyme is also known for its transgalactosylation activity and synthesis of lactose-based derivatives including lactulose and galactooligosaccharides, which can be used in probiotic foods.
