摘要

A method is described for the covalent attachment of DNA to asolid surface at high density for hybridization detec- tion by massspectrometry. A silicon wafer is function- alized to place an aminogroup on the surface; a hetero- bifunctional cross-linking agent isthen reacted with the primary amine to incorporate an iodoacetamidogroup. An oligodeoxynucleotide containing a 3'-or a 5'-disulfide istreated with a reducing agent, resulting in a terminal free thiol,which is then coupled to the iodoacetamido surface.
