One hundred sixty one fungal isolates were obtained from forty soil samples collected from various sites of Meerut region. On screening for phytase activity thirty three isolates were able to grow on medium containing Ca-phytate as sole source of phosphorus. Among 33 isolates, 15 were showed significant zone of hydrolysis. In submerged fermentation using phytase screening broth (PSB) as a production medium, isolate 15E gave highest titer of 31.76 U/mL among the isolates at pH 6.0. while in solid state fermentation using Rice bran and 1% maize flour as substrate a titre of 36.21 U/gDBM was achived at pH 6. The effect of temperature on phytase production among the isolate resulted in an enhancement of approximately 15% from 20 to 40°C. While the production decrease to -29 - 68 % at 60°C among the isolates. On altering the pH of production medium in submerged fermentation maximum phytase titer in production medium was observed at pH 4.0 and 6.0, with a very slight difference.
