Cotyledon and hypocotyl explants excised from 5-6-day-old in vitro-grown seeds of B. juncea cultivars RH30 and RH8812 were cultured on 8 MS media differing in their concentrations of growth regulators. Cultures were incubated at 25 +- 1degC with light/dark cycles of 16/8 h, and scored for percentage of explants regenerating shoots. For rooting, regenerated shoots were transferred to MSR (MS medium + 2 mg IBA/litre) or MS0 (MS without growth regulators). Highest regeneration frequency (80-90% from cotyledons, 37-40% from hypocotyls) was obtained on MS medium supplemented with 2.0 mg BAP [benzyladenine] and 0.2 mg IAA/litre. When IAA was replaced with NAA frequency of shoot regeneration decreased, but extensive rooting was observed.
