Amsonia orientalis Decne. (syn. Rhazya orientalis (Decne.) A. DC.) is an ornamental and medicinal plant which has restricted distribution only in the northwest of Turkey and the northeast of Greece. Nodal segments excised from 7-year-old plants were cultured in MS medium supplemented with 0.5, 1.0, 2.0, 4.0 mg l~(-1) BAP for shoot multiplication. Medium containing 1.0 mg l~(-1) BAP suggested regeneration of the highest number of shoots (9.2 ± 0.2 mean number of shoots at 91.1 ± 2.3% frequency). Nodes obtained from multiplied shoots were excised and cultured in MS medium supplemented with combinations of BAP, kinetin, IAA, and 2,4-D. Maximum mean shoot length of 4.4 ± 0.7 cm was noted from the medium supplemented with 0.5 mg l~(-1) IAA + 1.5 mgl~(-1) BAP and 0.5 mg l~(-1) IAA + 0.5 mgl~(-1) kinetin individually. Rooting experiments were also conducted in half- and full-strength MS medium supplemented with IAA or IBA. All shoots could be induced to root on full-strength MS medium supplementedwith 0.5 mg l~(-1) IAA within 4 weeks with a fairly good mean number and length of roots i.e. 7.9 ± 0.3 and 2.8 ± 0.2 cm. Plantlets went through a hardening phase in a controlled plant growth chamber prior to in vivo transfer. Eighty-four percent ofthe plantlets were succesfully acclimatized.
