Maintenanceofmitoticallycyclinggermlinestemcells(GSCs)isvitalforcontinuousproductionofgametes.Inwormsandinsects,signalingfromsurroundingsomaticcellsplayanessentialroleinthemaintenanceofGSCsbypreventingprematuredifferentiation.Inaddition,germcellproteinssuchastheemDrosophila/emPumilioandemCaenorhabditiselegans/emFBF,bothmembersofthePUFfamilytranslationalregulators,contributetoGSCmaintenance.FBFfunctionsbysuppressingGLD-1,whichpromotesmeioticentry.However,factorsthatdirectlypromoteGSCproliferation,ratherthanpreventdifferentiation,arenotknown.HereweshowthatPUF-8,anotheremC.elegans/emmemberofthePUFfamilyandMEX-3,aKHdomaintranslationalregulator,functionredundantlytopromoteGSCmitosis.WefindthatPUF-8proteinishighlyenrichedinmitoticgermcells,whichissimilartotheexpressionpatternofMEX-3describedearlier.Theempuf-8(minus;)mex-3(minus;)/emdoublemutantgonadscontainfarfewergermcellsthanbothsinglemutantsandwild-type.Whilethesecellslackmitotic,meioticandspermmarkers,theyretainthegermcell-specificPgranules,andarecapableofgametogenesisifGLP-1,whichnormallyblocksmeioticentry,isremoved.Significantly,wefindthatatleastoneofthesetwoproteinsisessentialforgermcellproliferationeveninmeioticentry-defectivemutants,whichotherwiseproducegermcelltumors.WeconcludePUF-8andMEX-3contributetoGSCmaintenancebypromotingmitoticproliferationratherthanbyblockingmeioticentry./p/div
