Objective: To study the relationship between osteopontin expression and cell invasion as wellas angiogenesis in preeclampsia placenta tissue. Methods: A total of 56 patients diagnosedwith preeclampsia in our hospital between June 2014 and March 2016 were selected aspreeclampsia group (PE group) and 80 normal subjects receiving antenatal examination andgiving birth during the same period were selected as control group. After labor, the placentatissue was collected, immunohistochemical staining kits were used to detect OPN expressionand enzyme-linked immunosorbent assay kits were used to determine cell invasion andangiogenesis molecule expression. Results: Positive OPN rate in placenta tissue of PE groupwas significantly lower than that of control group; cell invasion molecules LAMA4, uPA,Rho and ROCK as well as angiogenesis molecules VEGF, PIGF and AGF protein expressionin placenta tissue of PE group were significantly lower than those of control group, andcell invasion molecules LAMA4, uPA, Rho and ROCK as well as angiogenesis moleculesVEGF, PIGF and AGF protein expression in OPN-positive placenta tissue were significantlyhigher than those in OPN-negative placenta tissue. Conclusion: Lowly expressed OPNin preeclampsia placenta tissue will inhibit the expression of cell invasion molecules andangiogenesis molecules so as to inhibit sertoli cell invasion and the angiogenesis in placentatissue.
机译:目的:研究子痫前期胎盘组织中骨桥蛋白的表达与细胞侵袭以及血管新生的关系。方法:选择2014年6月至2016年3月在我院诊断为先兆子痫的患者56例作为先兆子痫组(PE组),同期选择80例接受产前检查和分娩的正常人作为对照组。产后收集胎盘组织,用免疫组织化学染色试剂盒检测OPN表达,用酶联免疫吸附试剂盒测定细胞侵袭和血管生成分子的表达。结果:PE组胎盘组织中OPN阳性率明显低于对照组。 PE组胎盘组织中细胞侵袭分子LAMA4,uPA,Rho和ROCK以及血管生成分子VEGF,PIGF和AGF蛋白的表达均明显低于对照组,而细胞侵袭分子LAMA4,uPA,Rho和ROCK以及血管生成分子OPN阳性胎盘组织中VEGF,PIGF和AGF蛋白的表达明显高于OPN阴性胎盘组织。结论:子痫前期胎盘组织中低表达的OPN会抑制细胞侵袭分子和血管生成分子的表达,从而抑制sertoli细胞侵袭和胎盘组织中的血管生成。
