Inthisstudy,itwasaimedtoinvestigatethemorphologiceffectsofsodiumbenzoate(SB)andcitricacid(CA),additivesusedinfood,onratliverandlivertissuesandchangesintotalsialicacid(TSA)levels.Atotalof30Wistaralbinoratsweighing200-250grweredividedto3groupswith10ineach.Ratsincontrolgroup,SBgroupandCAgroupwereadministeredonbygavagetapwater,Na-benzoate(2442mg/kgbodyweight)andcitricacid(576mg/kgbodyweight)oncedailyrespectivelyfor10days.Attheendoftheexperiment,animalsincontrolandexperimentgroupswereopenedandheartblood,hepaticandrenaltissueswereobtained.Removedtissueswereexaminedunderlightandelectronmicroscopyfollowingroutinefollowupprocedures.TSAamountsinserumandtissuesweremeasuredwithspectrophotometricmethod.InSBandCAgroups,disruptionwasobservedinhepatictissueengineering,vacuolizationwasobservedinhepatocytesandalsopyknoticnuclei,nucleuslossesandhypertrophywereobserved.InSBandCAgroups,pyknoticnucleiwereobservedinrenaltubularcells,membraneinjurywasobservedinapicalandbasalsurfacesoftubularcells,degenerationwasobservedinglomerularstructuresandirregularitywasseenintubularstructures.Plasmarhexis,lossesinmitochondrialcrista,degenerationinoutermembraneandnuclearmembraneofmitochondria,vacuolizationinhepatocytesandmyelinfigureswereobservedelectromicroscopicallyduetoSBandCA.Inrenaltissues,electronmicroscopicdegenerativechangesincludedelectron-densegranules,cristalossesinmitochondria,irregularmicrovilli,vacuolizationandplasmarhexisincytoplasmaoftubularcells,irregularityinchromatinmaterial,disruptioninbasalmembrane,enlargementinERsacs.InSBgroups,serumTSAamountwasseentodecreasesignificantly(p0.05)andasignificantdifferencewasnotseeninCAgroup.ItwasseenthatsialicacidamountssignificantlydecreasedbothinliverandkidneytissuesinSBgroup(p0.0001);inCAgroup,whileasignificantdifferencewasnotseeninhepatictissues,renalTSAlevelwasdetectedtoincreasesignificantly(p0.0001).WheneffectsofSBandCAonTSAamountandmorphologicdegenerationsinliverandrenalkidneywereevaluatedtogether,weconsiderthatmoredetailedstudiesarerequiredabouttheuseofthesefoodpreservatives.
