In this paper, we present a method for the detection of antibiotic resistance genes belonging to thediverse group of -lactamases based on real-time monitoring of a rolling-circle amplification (RCA),4 and a simple electrochemical sensor for capacitatively coupled contactless conductivity (C )measurements. Impedance spectra are presented to describe the underlying principles of the sensor andto find optimum operating conditions. Conductivity changes induced by the formation of amplificationproducts were monitored. By comparing the time responses we were able to distinguish amplificationreactions starting with DNA target concentrations as low as 0.42 nM from negative controls containingno target DNA.
