Toexaminetheeffectsofbuffaloratliver(BRL)cellsonthepreimplantationdevelopmentofmouseembryosinvitro,wefirstculturedtwo-cellmouseembryosaloneinserum-freeDulbeccomodifiedEaglemedium.Asexpected,theembryosdidnotdeveloptosubsequentstages.However,whencoculturedwithBRLcells,theembryosdevelopedtotheblastocyststageefficiently.DirectcontactofembryoswithBRLcellswasnotnecessaryfordevelopment:themediumconditionedbyBRLcellscontainedsolublefactorsthatsupportedthepreimplantationdevelopmentofmouseembryos.EmbryosculturedwithBRL-conditionedmediumthatwasreplacedatvariousintervalshadafurtherincreasedrateofdevelopmenttotheblastocyststage.Thisfindingindicatedthattheactivitiesofthefactorsweremaintainedonlybriefly.Sevenproteinsbetween35and44kDathatweredetectedinthemediumwerehighlybeneficialtothedevelopmentoftheembryos.Follistatin-relatedproteinandpigmentepithelium-derivedfactorarebelievedtobethefactorssupportingembryodevelopment.Theotherfiveproteinsalsomayimprovetheenvironmentforthedevelopmentofmouseembryosculturedinvitro.
