Determination of Vanillin, Eugenol and Isoeugenol by RP-HPLC

A sensitive RP-HPLC method with ultra-violet (UV) detection has been developed for the simultaneous determination of vanillin, eugenol and isoeugenol in bioconversion broth. Sample preparation was performed by filtration with ethyl alcohol added. Analysis of sample was performed on a Lichrospher 100 RP-18 column with a gradient elution of a mobile phase comprising methanol and acetic acid aqueous solution. The eluate was detected at 270 nm. Vanillin, eugenol and isoeugenol were eluted at 3.2 min, 10.7 min and 14.4 min respectively. Their standard curves were linear over the concentration ranges of 0–0.8 g L−1, 0–1.5 g L−1 and 0–1.0 g L−1; their limits of the detection (LODs) were 6.79 ng mL−1, 0.51 μg mL−1 and 0.099 μg mL−1; their recoveries were in the range of 95.3%–99.7%, 95.0%–102.8% and 96.4%–104.0% respectively.
