Activation of the Pacidamycin PacL Adenylation Domain by MbtH-like Proteins

Nonribosomal peptide synthetase (NRPS) as-nsembly lines are major avenues for the biosynthesis of anvast array of peptidyl natural products. Several hundrednbacterial NRPS gene clusters contain a small (∼70-residue)nprotein belonging to the MbtH family for which nonfunction has been defined. Here we show that two strictlynconserved Trp residues in MbtH-like proteins contributento stimulation of amino acid adenylation in some NRPSnmodules. We also demonstrate that adenylation can benstimulated not only by cognateMbtH-like proteins but alsonby homologues from disparate natural product pathways
