Instrumentation The chromatography was carried out using a Shimadzu system composed of SCL 10A VP Controller, LC 10AD pumps, SIL 10A Auto sampler injector, Prontosil 120-3-C18 ACE-EPS column oven, SPD M10AVP photodiode array detection (DAD) with a UV–Vis Detector system (all from Shimadzu, Kyoto Japan). Theobromine was separated on a reversed C18 column (150 × 4.6 mm; 3 µm id.; particle size, 3 mm) from BISCHOFF Analysentechnik und – geräte GmbH Leonberg Germany. The chromatography was carried out using Methanol–water 45:55 (v/v) as mobile phase at a flow rate of 0.8 ml/min and the run time was 20 min. The column temperature was 30 °C and the analyte was detected at 220 nm.
