不同营养和培养条件对向日葵茎溃疡病菌生长特性的影响

             

摘要

[目的]研究向日葵茎溃疡病菌(Phomopsis helianthi)生态适应性中的营养和培养条件,以期建立常规生物学检测方法,以便有效采取检疫处理措施.[方法]不同营养和培养条件下(包括培养基、碳源、氮源、温度、酸碱度和光照),对该菌培养7d,测量其菌落直径大小和观察其生长状况.并用SPSS 12.0软件进行相应的计算处理、统计分析.[结果]向日葵茎溃疡病菌在PDA、CMA培养基上生长较好,在改良的查彼培养基和WA培养基上生长较慢.该病菌能充分利用酵母膏和硝酸钾.在6~34℃均能生长,最适生长温度为23~26℃.适宜生长pH 3.0~12.0,最适pH 5.0~10.0.在全光照培养条件下有助于菌丝的生长.[结论]通过不同营养和培养条件下的生长情况,可知该病菌的最适生长条件,有助于对该病菌进行有效的检疫处理.%[ Objective ] The nutrition and culture condition of ecological adaptability for phomops - is helianthi has been studied. The conventional biological detection method was established, in order to take effective quarantine treatment measures. [ Method ] The fungus has been cultivated 7 days in different nutrients and culture condition (including media, carbon sources, nitrogen source, temperature, acidity and alkalinity, light condition), and its colonies diameter was measured and the growth status observed. And software SPSS 12.0 was used for corresponding calculation processing and statistical analysis. [Result] The result showed that the fungus could grow better on the PDA and CMA than modified Czapek and WA. It could fully utilize the yeast extract and potassium nitrate. It could grow within 6 - 34℃, and 23 - 26℃ was its optimum temperature. The fungus could grow in pH 3.0 to pH 12.0, and its optimal pH was pH5. 0 to pH 10.0. Full light was beneficial to the mycelium growth of Phomopsis helianthi. [ Conclusion ] The growth of the fungus in different nutrients and culture condition has been studied. The optimum growth condition of fungus was known, which would help us to take some effectively quarantine treatment.
