不同营养和培养条件对向日葵黑茎病菌生长特性的影响

         

摘要

[Objective] Ihe nutrition and culture conditions of ecoIogical adaptability for Phoma heliarthi Taberosi have been Btudied to establish the conventional biological detection method in order to take effective quarantine treatment measures. [Method] Ihe fungus has been cultivatived for 7 days under different nutrition and culture conditions (including media, carbon sources, nitrogen source, temperature, acidity and alkalinity, light condition) , and its colonies diameter were measured and the growth status were observed .And software SPSS 12.0 was used for corresponding calculation processing and statistical analysis . [ Result]The resuh showed that the fungus could grow better on the PDA and HIA than on WA. It could fully utilize the starch soluble and potassium nitrate. It could grow within 4 - 32℃ , and 24 - 28℃ was its optimum temperature. The fungus could grow better in pH 4.0 to pH 8.0, and its optimal pH was pH 5.0 to pH 7.0. Full light was beneficial to the maycalial growth of Phoma heliarTHi Taberosi. [ Conclusion] The growth results of the fungus under different nutrition and culture conditions have been studied . The optimum growth conditions of fungus were made clear, which help us to take some effectively quarantine treatment.%[目的]对向日葵黑茎病菌Phoma helianthi Taberosi生态适应性中的营养和培养条件进行研究,以期建立常规生物学检测方法,以便采取有效检疫处理措施.[方法]不同营养和培养条件下(包括培养基、碳源、氮源、温度、酸碱度、光照),对该菌培养7 d,测量其菌落直径大小和观察其生长状况.并用SPSS 12.0软件进行相应的计算处理、统计分析.[结果]向日葵黑茎病菌在PDA、HLA培养基上生长较好,在WA培养基上生长较慢.该病菌能充分利用可溶性淀粉和硝酸钾.在温度4-32℃均能生长,最适生长温度为24-28℃,适宜生长的PH4.0-8.0,最适PH5.0-7.0;在全光照培养条件下有助于菌丝的生长.[结论]通过不同营养和培养条件下的生长情况,可知该病菌的最适生长条件,有助于对该病菌进行有效的检疫处理.
