生脉成骨胶囊在激素性股骨头坏死防治中的应用研究

         

摘要

目的:通过研究生脉成骨胶囊对激素性股骨头缺血性坏死的股骨和髂骨骨髓间充质细胞增殖及矿化结节生成作用,及骨髓间充质细胞的碱性磷酸酶(ALP)和骨钙素(BGP)的变化,探讨其对机体髂、股骨间充质细胞成骨能力的影响,了解生脉成骨胶囊在防治激素性股骨头坏死方面的作用机理.方法:将40只SD雄性大鼠随机分为4组:空白对照组、模型组、生脉成骨预防用药组、生脉成骨治疗组,通过测定ALP、BGP、充质细胞的增殖能力、矿化结节数,推断股骨和髂骨近端成骨能力变化规律.结果:空白对照组、生脉成骨预防用药组、生脉成骨治疗组股骨髂骨的ALP、BGP含量均显著高于模型组(P<0.05);各组股骨、髂骨间充质干细胞向成骨细胞转化增殖的吸光度值(OD570)和矿化结节数明显高于模型组;生脉成骨预防用药组股骨和髂骨矿化结节数、ALP高于生脉成骨治疗组.结论:生脉成骨胶囊可以促进激素性股骨头坏死大鼠模型的股骨、髂骨间充质细胞碱性磷酸酶和骨钙素合成及骨髓间充质细胞向成骨细胞转化的细胞增殖及矿化结节形成.认为生脉成骨胶囊防治激素性股骨头缺血性坏死的作用机理与促进成骨细胞的分化成熟程度以及提高成骨细胞骨蛋白的分泌速度有关.%The influences of ShengMai ChengGu capsule on the formation of mesenchymal cell in iliac bone and femur were explored and the mechanism of ShengMai ChengGu capsule in prevention of steroid-induced necrosis of femoral head was understood through studying the function of ShengMai ChengGu capsule on the proliferation of mesenchymal cell and mineralization nodules in ischemic necrosis of iliac bone and femur caused by steroid and the changes of alkaline phosphatase (ALP) and bone gla protein (BGP) of mesenchymal cell. Method: 40 SD male rats were randomized into control group, model group, preventive group of ShengMai ChengGu and ShengMai ChengGu group. ALP, BGP, proliferation and mineralization nodules of mesenchymal cell were detected to conclude the rules of bone formation of proximal portions of iliac bone and femur. Results: The contents of BGP and ALP in femur and iliac bone of control group, preventive group of ShengMai ChengGu, ShengMai ChengGu group were remarkable higher than that of model group (P<0.05). The numbers of OD570 of mesenchymal cell transforming into osteoblast and mineralization nodules in femur and iliac bone of each group were obviously higher than that of model group; ALP and mineralization nodules in preventive group of ShengMai ChengGu were higher than that of ShengMai ChengGu capsule group. Conclusion: ShengMai ChengGu capsule can promote the composition of ALP and BGP in mesenchymal cell of femur and iliac bone in rat model with steroid-induced necrosis of femoral head as well as proliferation of mesenchymal cell transforming into osteoblast and mineralization nodules. The mechanism of ShengMai ChengGu capsulein prevention steroid-induced necrosis of femoral head is initially considered to be related to promoting osteoblast differentiation mature degree and raising osteoblast protein secretion speed.
