姜黄素抑制乳腺癌细胞株Survivin表达及其增殖的影响

摘要

Objective:To explore the effects of curcumin on Survivin expressions and the proliferation in breast cancer cell line MCF-7.Methods:CCK-8 method was used to determine the effects of curcumin on MCF-7 cells proliferation;real-time PCR and Western blot method were adopted to measure the expressions of Survivin mRNA and protein before and after curcumin acting on MCF-7 cells.Results:Curcumin could significantly inhibit the profiferation of breast cancer cell line MCF-7,and they showed concentration dependent,IC50 was 6.4 μM after dealing with curcumin.Meanwhile the expressions of Survivin mRNA and protein decreased notably (P＜0.05),the decrease of 20μM group was more notable compared with 10 μM group (P＜0.05).Conclusion:Curcumin could inhibit the proliferation of breast cancer cell line MCF-7,and its mechanism might be related to down regulating Survivin expressions.%目的:探讨姜黄素对乳腺癌细胞株MCF-7中Survivin表达及其增殖的影响.方法:采用CCK-8法检测姜黄素对MCF-7细胞增殖的影响;real-time PCR、Western blot检测姜黄素作用于MCF-7细胞前后,Survivin在mRNA和蛋白水平的表达.结果:姜黄素可显著抑制乳腺癌细胞株MCF-7的增殖,并呈浓度依赖性,姜黄素处理48小时的IC50值为6.4μM.同时Survivine在基因及蛋白的水平表达均显著降低(P＜0.05),20μM组较10μM组下降明显(P＜0.05).结论:姜黄素可以抑制乳腺癌细胞株MCF-7的增殖,其机制可能与下调Survivin表达相关.
