颠茄合剂中托烷类生物碱的定性与定量研究

         

摘要

ABSTRACT:OBJECTIVE To establish a method for qualitative and quantitative determination of the trapane al-kaloids in Inbelladonna mixtuxe.METHODS RP-HPLC methods was adopted for the atropine , scopolamine and anisodamine identification and the content of Atropine sulfate was also determined.The analyses were performed on a ZORBAX SB-C18(4.6 ×150mm,5μm) column with the mobile phase consisted of acetonitrile-phosphate buffer (pH 2.8)(13∶87)at a flow rate of 1.0mL· min-1,the detection wavelength was 210nm,the temperature of column was 30℃,and the injection volume was 20μL.RESULTS The chromatogram of the sample solution showed similarity to the reference solution.The linear range of Atropine sulfate was in the range of 15.06 ~120.5μg · mL -1 ( r =0.9998),the average recovery(n=9) was 98.27%and the RSD was 0.85%.CONCLUSION The method is ac-curate,reliable and specific ,which can be used for the quality contorl of Belladonna mixture.%目的:对颠茄合剂中托烷类生物碱进行定性与定量研究。方法利用反相高效液相色谱法对颠茄合剂中硫酸阿托品、东莨菪碱及山莨菪碱进行定性鉴别,同时对硫酸阿托品进行含量测定。色谱条件:色谱柱:ZORBAX SB-C18(4.6×150mm,5μm);流动相:乙腈-磷酸盐缓冲液(取磷酸二氢钾6.8g溶于1000mL水中,加三乙胺10mL,调pH至2.8)(13∶87),检测波长:210nm;流速为1mL· min -1,进样量20μL,柱温:30℃。结果样品溶液色谱图中显示与对照品溶液色谱图中相同的色谱峰。硫酸阿托品在15.06~120.5μg · mL -1范围内呈良好线性关系( r =0.9998);平均回收率(n=9)为98.27%,RSD为0.85%。结论该方法准确、可靠、专属性强,为建立颠茄合剂质量标准提供依据。
