朱红橘及其突变体牛肉红金橘的AFLP分析

         

摘要

为弄清朱红橘红色突变体牛肉红金橘遗传的真实性,利用AFLP分子标记技术对朱红橘(Citrus reticulata Blanco)及其实生变异体牛肉红金橘进行了遗传鉴定.结果表明:从120对EcoRI/MseⅠ酶切引物中筛选出具有多态性的引物7对(MCO5E04,MCO4E01,MC04E02,MC04E03,MC04E04,MC04E06,MCO5E02);对朱红橘及其突变体牛肉红金橘进行AFLP扩增,共扩增出648条带,其中,72条具有多态性,多态性比例占11.11%;应用NTSYSpc 2.1软件统计分析,品种(系)间的遗传距离为0.0215853.牛肉红金橘与朱红橘在DNA水平上确实存在差异,牛肉红金橘为遗传上稳定的变异体,具有成为新的红肉品种的遗传基础,可作为新型材料加以保护和利用.%AFLP technique was used to determine DNA polymorphiam of Zhuhongju and its variant line Niurou Hongjinju ( Citrus reticulata Blanco) to study the genetie diversity of Zhuhongju ( Citrus neticulata Blanco) and its red variant line. The results showed that seven primer pairs( MC05-E04 .MCO4-E01 , MC04-E02 . MC04-E03 . MC04-E04 , MC04-E06 , MC05 -E02 ) were selected out of 120 pairs of primer combinations and 648 polymorphism bands were amplified , 72 of which were polymorphic. Ratio of polymorphic hands was 11. 11 % . The genetic diatance between ZhuhongjU and its variant line was 0. 0215853 by NTSYSpe 2. 1 cluster analysis. Niurouhongjinju was different with Zhuhongju at DNA level and was a genetically stable variant which had got genetic basis to be seleted as a new cultivar and thould be protected and utilized as a new material.
