维生素D受体在乳腺癌进展过程中mRNA及蛋白表达情况及其意义

         

摘要

目的:通过测定维生素D受体( VDR)在乳腺疾病进展过程中的mRNA及蛋白表达情况,以探讨VDR在乳腺疾病演变过程中的表达变化及其意义。方法成组设计,分为四组(非特殊类型浸润性癌、正常乳腺、腺病、导管原位癌),采用逆转录实时荧光PCR( Real-time RT-PCR)方法检测乳腺标本的VDRmRNA的表达情况,采用免疫组织化学染色方法检测VDR的蛋白水平表达情况。结果在浸润性乳腺癌组织中,VDRmRNA表达水平显著降低,与正常乳腺组织相比差异有统计学意义(P<0.05),同时蛋白表达水平降低;正常乳腺组、腺病组与导管原位癌组VDRmRNA表达水平及蛋白水平依次降低,但差异无统计学意义(P>0.05)。结论浸润性乳腺癌组织中VDR表达下调,在本实验中显示VDR的表达与乳腺病变的进展呈负相关,进一步佐证VDR的表达对于乳腺有一定保护作用。%Objective To measure the expreassion of VDR mRNA and protein in the human mammary,and discuss the significance of VDR in the etiology of breast cancer. Methods Using all patients were divided into 4 groups:non special type in-vasive carcinoma、normal breast、adenosis、ductal carcinoma in situ ,Real-time RT-PCR method and immunohistochemical method were used to examine the expreassion of VDR mRNA and protein in each group. Result In non special type invasive carcinoma group ,the level of VDR mRNA was significantly decreased, compared with the normal breast group ,which had obviously statis-tical significance (P<0. 05),accompanying with the expreassion of VDR protein;the expreassion of VDR mRNA and protein in normal breast、adenosis、ductal carcinoma in situ were decreased successively,which had no obviously statistical significance( P>0. 05) . Conclusion The expreassion of VDR was down-regulated in non special type invasive carcinoma redated with the malig-nancy in breast diseases. VDR had a profitable function in preventing the progression of breast cancer.
