丁香酚急性毒性和皮肤刺激试验∗

摘要

Objective:To evaluate the acute toxicity and skin irritation of Eugenol.Methods:SD rats were administered by oral ga-vage with eugenol at a dose of 5250,4200,3360,2680 and 2149 mg·kg-1 for once,respectively.After 2 weeks of clinical observa-tion,the LD50 of eugenol on rats was determined by Bliss method.Eugenol was daubed on the intact skin of rabbit at a concentration of 1060 and 265 mg·ml-1 to test whether it may induce stimulus.Results:The LD50 of eugenol on rats administered by oral gavage was 3189.9 mg·kg-1 ,and the 95% confidence intervals were (2423.1 ~ 3711.8)mg·kg-1 .1060 mg·ml-1 of eugenol showed moderate irritation to rabbit skin,and this adverse effect was reduced with the decreasing concentration of the Eugenol.Conclusion:Skin irritation effects of eugenol decrease with concentration.%目的：：对丁香酚进行大鼠灌胃急性毒性试验和兔完整皮肤给药刺激性试验,了解其安全性。方法：丁香酚分别以5250,4200,3360,2680和2149 mg·kg-1剂量对大鼠灌胃给药,观察2周,用 Bliss 法测定大鼠的 LD50；以1060 mg·ml-1和265 mg·ml-1单次涂抹兔完整皮肤,观察兔皮肤的刺激性反应。结果：丁香酚大鼠灌胃的 LD50为3189.9 mg·kg-1,95%可信限为(2423.1~3711.8)mg·kg-1,1060 mg·ml-1对兔完整皮肤有中度的刺激性,随浓度降低刺激性减弱。结论：丁香酚的皮肤刺激性随浓度降低而减弱。
