Gab2的表达及其对胃癌细胞增殖的影响

摘要

目的：探讨 Gab2在胃癌中的表达及其对胃癌细胞增殖的影响。方法应用免疫组织化学的方法检测65例胃腺癌中 Gab2的表达，将含 Gab2-siRNA 的载体质粒转染胃癌细胞 BGC-823，用蛋白印迹法检测胃癌细胞中 Gab2蛋白表达水平的变化，用噻唑蓝（MTT）法检测肿瘤细胞增殖情况，末端脱氧核苷酸转移酶介导的dUTP 缺口末端标记测定法（TUNEL）检测细胞凋亡情况。结果Gab2在胃腺癌中过表达，阳性表达率为69％，而在正常胃黏膜中低表达，转染 Gab2-siRNA 后，Gab2蛋白表达水平显著降低，肿瘤细胞的增殖受到抑制，细胞凋亡率达到42．34％（P ＜0．05）。结论Gab2在胃腺癌中过表达，靶向 Gab2的 RNA 干扰能够有效地抑制胃癌细胞的增殖，并促进其凋亡，因此其可能在胃癌的发生发展中起着重要作用。%Objective To investigate the expression of Gab2 in the gastric cancer and to explore its relation-ship with gastric cancer cell proliferation.Methods The expression of Gab2 in the gastric cancer was measured by immnohistochemistry.Recombinant plasmid Gab2 was transfected into BGC-823 cells.The protein levels of Gab2 were detected by Western blot.The proliferation of BGC-823 cells was detected by MTT assay,and the apoptosis was detected by TUNEL assay.Results The positive expression rate of Gab2 in tumor tissues was 69%,signifi-cantly higher than that in normal gastric epithelium.After transfection of Gab2-siRNA,the protein levels of Gab2 was obviously reduced and the proliferation of BGC-823 cells was inhibited. The apoptosis rate of BGC-823 cells was 42.34%(P <0.05).Conclusion Gab2 is overexpressed in gastric cancer tissues,RNA interference of Gab2 inhibits the proliferation and induces the apoptosis of BGC-823 cells,so it may be involved in gastric cancer pro-gression.
