骨髓增生异常综合征的免疫表型和SURVIVIN、P15INK4B、TRF1基因表达

         

摘要

目的 探讨骨髓增生异常综合征(MDS)特征性免疫表型,SURVIVIN、P15INK4B、TRF1基因表达程度,及其与临床病情进展的关系.方法 抽取50例MDS患者骨髓细胞,用流式细胞学技术检测免疫表型;抽取单个核细胞,用RT-PCR方法检测SURVIVIN、P15INK4B、TRF1基因mRNA表达程度.结果 与对照组相比,MDS患者骨髓细胞原始群CD7和CD34表达增多;粒细胞群CD34表达增多,CD13、CD33、CD11b表达下降;单核细胞表达CD117和CD34.与对照组相比,MDS患者骨髓单个核细胞SURVIVIN和TRF1基因mRNA表达增加,P15INK4B基因mRNA表达下降(P均<0.05).从低危组到高危组,SURVIVIN和TRF1基因mRNA表达增加,P15INK4B基因mRNA表达下降(P均<0.05).结论 MDS免疫表型有其特征性,SURVIVIN、P15INK4B和TRF1基因表达可能与其发病进展有关.%Objective To explore the characteristic immunophenotype and SURVIVIN, P15INK4B and TRF1 gene of myelodysplastic syndromes (MDS), and the its relation with the progression of clinical condition. Methods Immunophenotype analysis was performed in 50 cases of MDS using flow cytometry (FCM ). We evaluated the mRNA expression of SURVIVIN, P15INK4B and TRF1 in mononuclear cells of MDS bone marrow by reverse transcription-polymerase chain reaction (RT-PCR). Results Compared with the control group, CD7 and CD34 expressions in primary cell populations of bone marrow cell, CD34 expression in granule cell were increased, CD13, CD33, CD11b decreased in MDS patients. CD117 and CD34 expressed in monocytes. Compared with the control group, P15INK4B mRNA was decreased, but SURVIVIN and TRF1 increased. From low-risk group to the high-risk group, SURVIVIN and TRF1 mRNA expressions were increased, but P1SINK4B decreased gradually. Conclusions Immunophenotype of MDS has its characteristic. Progress and incidence of MDS may have relation with SURVIVIN, P15INK4B, and TRF1 gene.
