缬沙坦对野百合碱诱导肺动脉高压模型大鼠血浆CRP、ET-1和NO水平的影响

     

摘要

Objective To observe the changes of c-reactive protein (CRP), endothlin (ET) and nitric oxide (NO)in pulmonary hypertensive rats induced by monocrotaline after valsartan interventions. Methods The healthy Wistar rats were injected with 60 mg/kg of monocrotaline, after 4 weeks, the models were reproduced. The intervention group was treated with 20 mg/( kg · d) of valsartan intragastrically for 4 weeks. The parameters of pulmonary hemodynamics were monitored, the levels of CRP, ET-1 and NO were detected. Result The levels of the mean pulmonary arterial pressure, CRP,ET-1 were declined and the level of NO was increased after valsartan interventions (P < 0.05 ). Conclusion Valsartan can decrease the mPAP and ameliorate the levels of CRP, ET-l/NO in pulmonary hypertensive rats, the mechanism maybe related with its effects of protecting endothelial cells.%目的 观察缬沙坦对野百合碱(MCT)所致肺动脉高压大鼠血浆CRP、ET-1和NO水平的影响.方法 将健康雄性Wistar大鼠一次性项背部注射MCT(60 mg/kg),4周后成模,治疗组用缬沙坦20 mg/(kg·d)灌胃,持续4周达实验终点;然后经微导管介入测定大鼠肺动脉平均压(mPAP);计算右室肥大指数;分别测定血浆CRP、ET-1和NO水平.结果 缬沙坦可有效降低MCT所致肺动脉高压大鼠mPAP、血浆CRP、ET-1水平和升高NO水平(P<0.05).结论 缬沙坦可有效降低肺动脉高压大鼠肺动脉压,改善血浆CRP、ET-1和NO水平,作用机制可能与其对肺血管内皮细胞的保护作用有关.
